RESUMO

Abstract Coxiella burnetii is a zoonotic agent transmitted mainly by small ruminants. In Brazil the disease has been classified as a notifiable disease since 2013, when human cases were reported. This study aimed to identify risk factors associated with the presence of anti- Coxiella burnetii antibodies in goats and sheep in a semiarid region of Northeastern Brazil. Sera of 412 goats and 403 sheep from municipality of Petrolina, Pernambuco, were examined by the Indirect Fluorescent Antibody Test (IFAT) against antigens of C. burnetii. Information about management variables (independent variables) that could be associated with the presence of the microorganism (dependent variables) were obtained from the supervisor of each farm. It was determined that 2.2% (9/412) of the goats and 2.1% (9/403) of the sheep had antibodies reactive to C. burnetii. The presence of anti-C. burnetii antibodies was associated with the dry area of the Sequeiro (a region in the northern part of the municipality of Petrolina) (P = 0.025), male sheep (P = 0.020), and intensive goat breeding (P = 0.005). This study therefore showed the presence of anti-C. burnetii antibodies in goat and sheep, confirming for the first time that this agent is likely circulating among goat herds in the Caatinga Biome, semi-arid of Brazil.


Resumo Coxiella burnetii é um agente zoonótico transmitido principalmente por pequenos ruminantes. No Brasil, a doença foi classificada como de notificação compulsória desde 2013, quando casos humanos foram relatados. O objetivo deste estudo foi identificar os fatores de risco associados à presença de anticorpos anti-Coxiella burnetii em caprinos e ovinos em uma região semiárida do Nordeste do Brasil. Este estudo envolveu um inquérito sorológico de 412 caprinos e 403 ovinos em fazendas do município de Petrolina, no estado de Pernambuco. Os soros foram examinados pela Reação de Imunofluorescência Indireta (RIFI) contra antígenos de C. burnetii . Informações sobre variáveis ​​de manejo (variáveis ​​independentes) que poderiam estar associadas à presença do microrganismo (variáveis ​​dependentes) foram obtidas do proprietário de cada fazenda. Foi determinado que 2,2% (9/412) dos caprinos e 2,1% (9/403) dos ovinos tinham anticorpos reativos a C. burnetii. A presença de anticorpos anti-C. burnetii foram associados com a área seca do Sequeiro (região no norte do município de Petrolina) (P = 0,025), ovinos machos (P = 0,020) e criação intensiva de caprinos (P = 0,005). Este estudo, portanto, observou a presença de anticorpos anti-C. burnetii em pequenos ruminantes, confirmando pela primeira vez que este agente pode estar circulando em rebanhos caprinos no bioma Caatinga, semiárido do Brasil.

Abstract Ovine/caprine ureaplasmas have not yet been assigned a species designation, but they have been classified into nine serotypes. Herein ureaplasmas were searched for in 120 samples of vulvo vaginal mucous from sheep and 98 samples from goats at 17 farms. In addition, semen samples were collected from 11 sheep and 23 goats. The recovered ureaplasma were from sheep and goats from animals without any reproductive disorder symptoms, but not all animals presented positive cultures. In sheep, 17 (68%) cultures of vulvovaginal mucous were positive for ureaplasma and 11 (27%) samples of semen presented positive cultures in animals with clinical signs of orchitis, balanoposthitis or low sperm motility. In goats four ureaplasma isolates were obtained from vulvovaginal mucus, but the semen samples were all negative. The isolates were submitted to Pulsed-field gel electrophoresis methodology and their 16S rRNA genes were sequenced. Fifty percent of ureaplasma recovered from sheep allowed for PFGE typing. Eleven isolates showed eight profiles genetically close to the bovine ureaplasmas. The 16S rRNA gene sequencing showed differences or similarities of isolates from sheep and goats, and the reference strains of bovine and human ureaplasma. Four clinical isolates from sheep were grouped separately. The studied ureaplasma isolates showed to be a diverse group of mollicutes.

Se realizó un estudio epidemiológico de brucelosis en 516 majadas caprinas o mixtas (caprinos/ovinos) de las 3 regiones agroecológicas de la provincia de Formosa, Argentina. Mediante las pruebas de aglutinación en placa con antígeno tamponado y de fijación del complemento en suero se estudiaron un total de 25.401 caprinos y 2.453 ovinos. Además, se realizaron cultivos bacteriológicos y PCR en muestras de leche de cabras de 3 majadas con brucelosis y abortos recientes. Se detectó brucelosis en 4 de los 9 departamentos de la provincia, la prevalencia global fue del 2 % y la intrapredial varió entre el 1 y el 40%. La proporción de majadas positivas fue del 3,6, el 12 y el 36 % para las regiones este, centro y oeste, respectivamente. Se aisló Brucella melitensis bv. 1 de cabras por primera vez en la provincia. La PCR amplificó fragmentos esperados de 827 pb correspondiente al gen omp2ab (Brucella spp.) y de 731 pb correspondiente al inserto IS711 (B. melitensis). La detección de anticuerpos en ovinos que cohabitan con caprinos sugiere que las infecciones habrían sido causadas por B. melitensis, lo que constituye un riesgo adicional para la salud pública. Los programas de control y erradicación de la brucelosis deberían considerar las majadas mixtas como una sola unidad epidemiológica. Los resultados indican que la brucelosis por B. melitensis bv. 1 es altamente endémica en las regiones centro y oeste de la provincia de Formosa.


An epidemiological study of brucellosis was carried out in 516 goats and mixed flocks (goat/sheep) from the three agro-ecological regions of Formosa province, Argentina. Serum samples from a total of 25401 goats and 2453 sheeps were analyzed using buffered plate agglutination test (BPAT) and complement fixation test (CFT). Bacteriological and PCR analyses on milk samples from goats in three flocks with a history of brucellosis and recent abortions were also performed. Brucellosis was detected in four of the nine departments of the province with an overall prevalence of 2 % and an intra-flock prevalence ranging between 1 % and 40 %. The proportion of infected flocks was 3.6 %, 12 % and 36 % for the eastern, central and western regions, respectively. Brucella melitensis bv. 1 was isolated efrom goats for the first time in the province. The expected fragments of 827 bp from the omp2ab gene (Brucella spp.) and 731 bp from the insert IS711 (B. melitensis) were amplified by PCR. Detection of antibodies by BPAT and FCT in sheep cohabiting with goats suggests that infections could have been caused by B. melitensis, posing an additional risk to public health. Control and eradication programs for brucellosis should consider mixed flocks as a single epidemiological unit. The results indicate that brucellosis by B. melitensis bv1 is highly endemic in the central and western regions of Formosa province.

Multiple-locus variable number-tandem repeat analysis (MLVA) of Mycobacterium avium subspecies paratuberculosis (MAP) isolates may contribute to the knowledge of strain diversity in Argentina. Although the diversity of MAP has been previously investigated in Argentina using IS900-RFLP, a small number of isolates were employed, and a low discriminative power was reached. The aim of the present study was to test the genetic diversity among MAP isolates using an MLVA approach based on 8 repetitive loci. We studied 97 isolates from cattle, goat and sheep and could describe 7 different patterns: INMV1, INMV2, INMV11, INMV13, INMV16, INMV33 and one incomplete pattern. INMV1 and INMV2 were the most frequent patterns, grouping 76.3% of the isolates. We were also able to demonstrate the coexistence of genotypes in herds and co-infection at the organism level. This study shows that all the patterns described are common to those described in Europe, suggesting an epidemiological link between the continents.

Anaplasma species are obligate intracellular pathogens that can cause tick-borne diseases in mammalian hosts. To date, very few studies of their occurrence in Korean native goats (Capra aegagrus hircus) have been reported. In the present study, we investigated Anaplasma infection of Korean native goats on Jeju Island, Republic of Korea, and performed phylogenetic analysis based on the 16S rRNA gene sequences. Our results showed that Anaplasma infection was found mostly in adult female goats. The phylogenetic tree revealed that the 7 sequences identified in Korean native goats could belong to Anaplasma sp. and were distinct from A. marginale, A. centrale, and A. ovis. The results indicated that the sequences identified to belong to Anaplasma were closely related to sequences isolated from goats in China and were clustered within the same group. To our knowledge, this is the first study to detect Anaplasma sp. infection in Korean native goats.

The present study reports the first isolation of Actinobacillus seminis from a goat in Brazil. A four-year-old Moxotó breeding goat in a flock of 70 goats and 65 sheep reared together in the county of Patos, semiarid region of Northeastern Brazil, showed clinical signs of unilateral orchitis and epididymitis. Diagnosis of A. seminis infection was confirmed by association of clinical findings, bacterial isolation and 16S rRNA gene sequencing. This result suggests that A. seminis may be an additional cause of infertility in goats, and that sheep may be the source of infection because the mixed farming system allows the contact between sheep and goats in the semiarid region of Northeastern Brazil.

Listeriosis is a disease primarily of ruminants caused by the Gram-positive bacterium Listeria monocytogenes. Ruminants either demonstrate manifestations of the encephalitic, septicemic, or reproductive form of listeriosis. The pathological and molecular findings with encephalitic listeriosis in a 5.5-month-old, male, mixed-breed goat and a 3-year-old Texel-crossed sheep from northern Paraná, Brazil are described. Clinically, the kid demonstrated circling, lateral protrusion of the tongue, head tilt, and convulsions; the ewe presented ataxia, motor incoordination, and lateral decumbency. Brainstem dysfunctions were diagnosed clinically and listeriosis was suspected. Necropsy performed on both animals did not reveal remarkable gross lesions; significant histopathological alterations were restricted to the brainstem (medulla oblongata; rhombencephalitis) and were characterized as meningoencephalitis that consisted of extensive mononuclear perivascular cuffings, neutrophilic and macrophagic microabscesses, and neuroparenchymal necrosis. PCR assay and direct sequencing, using genomic bacterial DNA derived from the brainstem of both animals, amplified the desired 174 base pairs length amplicon of the listeriolysin O gene of L. monocytogenes. Phylogenetic analyses demonstrated that the strains associated with rhombencephalitis during this study clustered with known strains of L. monocytogenes lineage I from diverse geographical locations and from cattle of the state of Paraná with encephalitic listeriosis. Consequently, these strains should be classified as L. monocytogenes lineage I. These results confirm the active participation of lineage I strains of L. monocytogenes in the etiopathogenesis of the brainstem dysfunctions observed during this study, probably represent the first characterization of small ruminant listeriosis by molecular techniques in Latin America, and suggest that ruminants within the state of Paraná were infected by the strains of the same lineage of L. monocytogenes.

The morphometry of bronchial associated lymphoid tissue (BALT) and its possible exploration as to the control of common viral and bacterial pneumonia and their co-infection in Sub-Saharan goats is scanty in literatures. This investigation utilizes archival tissues from study that involved 35 apparently healthy West Africa Dwarf goats which were divided into groups PPRV and PPRV+MH with 15 goats each while 5 goats served as control. PPRV goats were infected intratracheally with 1ml of pure cultured 106.5 TCID50 PPR Pestes des petit Ruminant virus (PPRV) grown in baby hamster kidney cell lines and PPRV+MH with 1 ml of PPRV and a week later 1ml of MH (Mannheimia hemolytica) A2. The BALT number, type and cellularity were determined using standard methods with the use of an image analyzer. Student t-test was used to test for significant differences. The results showed that the average number of BALT was significantly more in PPRV+MH infection than that of PPRV infection i.e. 6.20+/-3.63 for PPRV+MH and 4.11+/-2.92 for PPRV with more nodular types of BALT (8.01+/-1.83) than the aggregates type (4.3+/-1.50) while the average number of lymphocytes in the nodular, aggregate BALT significantly increased with 82.25+/-15.95, 56.00+/-7.28 in PPRV to 103.33+/-44.10, 66.00+/-7.93 in PPRV+MH respectively. There was an irregular pattern in the surface Area (SA) and Perimeter (P) of BALT following infections. The increase in average number of BALT and lymphocytes in PPRV+MH infection than that of PPRV infection showed that the development of BALT and the number of lymphocytes in BALT could be used to assess the mucosal immunity following intratracheal administration of PPRV and MH in goats. This is however the first study that described the morphometry of bronchial associated lymphoid tissue in experimental PPRV and PPRV+MH infection in Sub-Saharan goats.


Es escaso en la literatuta el conocimiento de la morfometría del tejido linfoide asociado a bronquios (BALT) y su posible exploración para el control de la neumonía viral común, pneumonia bacteriana y su co-infección en el ganado caprino subsahariano. Esta investigación utilizó tejidos archivados pertenecientes a 35 cabras enanas del oeste africano aparentemente sanas, que fueron divididas en grupos PPRV y PPRV+MH con 15 cabras cada uno, mientras que 5 cabras sirvieron como control. Las cabras PPRV fueron infectadas por vía intratraqueal con 1 ml de cultivo puro 106,5 TCID50 PPR virus Peste de Pequeños Rumiantes (PPRV) cultivados en líneas celulares renales de hámsters recien nacidos y PPRV+MH con 1 ml de PPRV y una semana después de 1 ml de MH (Mannheimia haemolytica) A2. El número de BALT, tipo y la celularidad se determinaron utilizando métodos estándar con el uso de un analizador de imágenes. Se utilizó la prueba t de Student para probar las diferencias significativas. Los resultados mostraron que el número promedio de BALT fue significativamente mayor en la infección PPRV+MH que la infección PPRV, es decir, 6,20+/-3,63 para PPRV+MH y 4,11+/-2,92 para PPRV con un BALT principalmente de tipo nodular (8,01+/-1,83) que de tipo agregado (4,3+/-1,50), mientras que el número promedio de linfocitos en el BALT nodular y agregado aumentó significativamente con 82,25+/-15,95 y 56,00+/-7,28 en PPRV y, 103,33+/-44,10 y 66,00+/-7,93 en PPRV+MH respectivamente. No hubo un patrón irregular en el área de superficie y perímetro del BALT después de las infecciones. El aumento en el número promedio de BALT y linfocitos en la infección PPRV+MH comparado con la de infección PPRV mostró que el desarrollo de BALT y el número de linfocitos en BALT podría ser utilizado para evaluar la inmunidad de la mucosa después de la administración intratraqueal de PPRV+MH en cabras. The study into the pattern of distribution of the lung consolidation associated with common viral and bacterial pneumonia and their co-infection in subsaharan goats is scanty in literatures. Fifty apparently healthy West Africa Dwarf goats (WAD) six months of age were used for the experiment. The animals were divided into groups A, B, and C with 15 goats each while 5 goats served as control. Group A goats infected with 1ml of pure culture (1 X 109 CFU) of Mannheimia haemolytica MH A2, while group B with 1ml of pure cultured 106.5 TCID50 PPR virus grown in Baby hamster kidney cell lines and group C with 1 ml of PPRV and a week later 1ml of MH A2. The degree of consolidation or pneumonia as a percentage of the total lung volume was determined by visual observation, palpation and measurement of the lesion which is estimated as a percentage of each lobe. Student t-test were used to test for significant differences. The right lungs have a higher lung consolidation percentage than the left in all the treatment groups. The accessory lobe was affected in the PPRV group. The MH group has the highest lung consolidation percentage (10.1 percent). The PPRV 1-28dpi has the lowest consolidation percentage (1.06 percent). There is significant difference in the consolidation percentage and mortality between MH, PPR+MH, PPRV 28-45 dpi and PPRV 1-28dpi (P<0.05). This observation further show that the right lung and the anterior lobes were more affected in experimental viral and bacterial respiratory pathogen and their co-infection as the trachea birfucation is first to the right and the distance between the right and the left birfucation was 1.5 +/- 0.35cm. It is the first study that describes and compare the pattern of distribution and morphometry of pneumonia in experimental PPRV, MH and PPRV+MH infections in goats.


El estudio sobre el patrón de distribución de la consolidación pulmonar asociada con neumonía virales y bacterianas comunes y sus co-infección en cabras Subsaharianas, es escasa en la literatura. Cincuenta cabras enanas de África occidental (WAD) aparentemente sanas de seis meses de edad fueron utilizados para el experimento. Los animales se dividieron en grupos A, B y C con 15 cabras cada uno mientras que el 5 cabras sirvió como control. Grupo A cabras infectadas con 1 ml de cultivo puro (1 X 109 UFC) de Mannheimia haemolytica MH A2, mientras que el grupo B con 1 ml de cultivo puro 10 6,5 DICT50 PPR cultivado en líneas celulares de riñón de crías de hámsters y el grupo C con 1 ml de PPRV y un semana después de 1 ml de MH A2. El grado de consolidación o neumonía como porcentaje del volumen pulmonar total se determinó por observación visual, palpación y la medición de la lesión que se estima como un porcentaje de cada lóbulo. El test t de Student se utilizaron para probar las diferencias significativas. El pulmón derecho tiene un porcentaje de consolidación pulmonar superior a izquierdo en todos los grupos de tratamiento. El lóbulo accesorio se vio afectado en el grupo de PPRV. El grupo MH tiene el porcentaje más alto de consolidación pulmonar (10,1 por ciento). El PPRV 1-28dpi tiene el menor porcentaje de consolidación (1,06 por ciento). No hay diferencia significativa en el porcentaje de consolidación y la mortalidad entre MH, MH + PPR, PPRV 28-45 dpi y PPRV 1-28dpi (P <0,05). Esta observación muestra además que el pulmón derecho y los lóbulos anteriores se vieron más afectados en infecciones respiratorias patógenas experimentales con agentes virales y bacterianos y su co-infección como la bifurcación traqueal es primero a la derecha y la distancia entre la derecha y la bifurcación izquierda fue de 1,5 +/- 0,35 cm. Es el primer estudio que describe y compara el patrón de distribución y la morfometría de las neumonías en PPRV experimentales, MH y MH + PPRV...

O leite e derivados são reconhecidos como veículos de patógenos para humanos, secundário a contaminação pós-ordenha ou de infecções do próprio animal, particularmente na mastite. Foi estudada a ocorrência de mastite e aspectos do manejo em cabras de três propriedades criadas em sistema orgânico. O exame clínico da glândula mamária de 64 cabras em diferentes períodos de lactação, não acusou a presença de mastite clínica. Entretanto, o Califórnia Mastitis Test (CMT) identificou 54 (22,7 por cento) metades mamárias reagentes (+ ou ++). Foram colhidas 238 amostras de leite, das quais houve isolamento bacteriano em 37 (15,6 por cento). Em apenas oito amostras houve coincidência entre o isolamento bacteriano e o resultado do CMT, indicando sensibilidade de 21,6 por cento para este teste no diagnóstico de mastite subclínica em caprinos. Staphylococcus coagulase negativa (SCN) foi o microrganismo mais freqüente (83,8 por cento). O teste de sensibilidade microbiana in vitro revelou resistência das linhagens de SCN ao cotrimoxazol (50 por cento), ampicilina (48,1 por cento), nitrofurantoína (7,7 por cento), cefaclor (7,14 por cento) e oxacilina (3,85 por cento). Cefalotina, gentamicina, neomicina, estreptomicina e tetraciclina foram os antimicrobianos mais efetivos frente aos isolados. Não se evidenciou relação entre a ocorrência de mastite subclínica com a raça, a fase de lactação, sistema de ordenha ou qualidade da água utilizada nas propriedades.


Milk and derivates are recognized as vehicle of different pathogens for humans. These micoorganisms may manifest as post-milking contamination or as in infections in the animal itself, particularly in mastitis. The aim of present study was characterizes the occurrence of mastitis and management aspects in dairy-goats raised in three farms of organic milk. The correlation between mastitis and goat husbandry was also investigated. The clinical exam of the mammary glands of 64 dairy-goats did not reveal the presence of clinical mastitis. Nevertheless, the California Mastitis Test (CMT) identified 54 (22.7 percent) reactive mammary halves (+ or ++). From 238 milk samples collected, 37 (15.6 percent) were positive for bacterial isolates. Coincident positive results for bacterial isolate tests and CMT were observed in only eight samples, which indicate a sensitivity index of 21.6 percent for the caprine subclinical mastitis diagnosis test. Coagulase-negative staphylococci (CNS) were identified as the most frequent pathogen in milk samples (83.8 percent). The in vitro sensitivity test revealed CNS strains resistant to cotrimoxazole (50 percent), ampicillin (48.1 percent), nitrofurantoine (7.7 percent), cefaclor (7.14 percent), and oxacilline (3.85 percent). Cefalotine, gentamicin, neomycin, streptomycin, and tetracycline were the most effective drugs. No correlation was observed between the occurrence of subclinical mastitis and race, lactation period, drinking water quality in farms, or milking system as adopted in the present study.

Contagious Caprine Pleuropneumonia [CCPP] is one of the common infections in the middle east regions. So far, there has not been received any report about isolation and identification of these agents in Iran. The aim of this study is to diagnose and isolate mycoplasma agents in suspected goat flocks. Total of 100 pneumonic lung specimen from 20 CCPPsuspected flocks were collected from abbatoirs close to Kermanshah during 1384-1386 and had been sent to Microbiology Lab. Gross lesions showed hepatization with grey and white lesions [consolidation] and motley appearance with or without fibrin. The minced tissue were inoculated to PPLO broth agar. After multiple passages, typical mycoplasma colony was isolated from 4 flocks [22/2%]. Mycoplasma DNA was also extracted based on phenolchloroform method and subjected to generic PCR with specific primers. In addition to the perivious positive samples from tissue culture, 5other flocks also showed contamination with Mycoplasma organisms in PCR tests[45%]. Then, the samples were determined for Mycoplasma mycoides cluster infection, M. capricolum capripneumonia and M. mycoides mycoides [L.C], using M. agalactia as negative control, with specific primers in PCR, there has showed no contamination to these strains. However, to declare "free status " from CCPP in goat flocks requires more developed researches and much more samples in further investigation

In this study, we have tested the possible use of propolis, one of the few natural remedies that exhibits potential therapeutics properties, in combination with some antibacterial drugs for treatment of clinical ovine and caprine listeriosis that was described and diagnosed. Three private sheep flocks of 485 sheep and 2 private goat flocks of 275 goats were carefully clinically examined after appearance of different forms of suspected clinical listeriosis. The clinical diagnosis was confirmed by histopathological and bacteriological examinations. The infected sheep and goats were divided into groups based on the clinical form of listeriosis for field treatment. In ovine listeriosis, 73 out of 485 sheep were affected with 15.05% overall morbidity and 10.96% case fatality rates, respectively. In eaprine listeriosis, 55 out of 275 goats were affected with 20% overall morbidity rate and no case fatality rate. Encephalitic listeriosis was more prevalent among sheep while listerial abortion and septicaemic listeriosis were more prevalent among goats. Histopathologically, the brain tissues showed extensive perivascular mononuclear cuffing with microabscesses. Bacteriologicailly, L. monocytogenes was isolated from [31.09%] and [26 .9 8%] of collected samples in sheep and goats respectively. The field treatment revealed that the affected sheep and goats treated with biopropolis alone or with antibiotic combination regimen reduced the overall severity of listeriosis better than those treated with antibiotic alone [ampicillin or cefotaxime alone] or cefotaxime alternative with gentamicin and trimethoprimsuiphadimethoxine combination. Combined treatment with ampicillin-biopropolis dosage regimen was found to be the most effective

RESUMO

We describe the first report of caprine arthritis-encephalitis virus (CAEV) isolation in Brazil. In December 1989 a four-year old anglonubian arthritic doe was submitted to euthanasia and its synovial membranes were cultured by explantation. This doe was positive for antibodies to the caprine arthritis-encephalitis virus (CAEV). The reverse transcriptase activity detected in culture supernatants, the characteristic cytopathic effect of lentivirus in these cultures, the detection of viral antigens in concentrated supernatants by the agar gel immunodiffusion test, and the results of the fluorescent antibody test and of Northern blot analyses are consistent with the isolation of the CAEV
